EKSPLORASI MEKANISME MOLEKULER SEDIAAN IMUNOSTIMULAN DARI DAUN SUNGKAI TERHADAP EKSPRESI PERFORIN, GRANZIM, DAN INTERFERON GAMMA DALAM SISTEM IN VIVO

Sungkai leaf plant (Paronema canescens Jack.) is a plant that is spread in Southeast Asia such as in the forests of Sumatra and Borneo, Indonesia. This plant is ethnobotanically widely used by the community in treating various diseases. This is thought to be due to the high content of secondary metabolite compounds in the plant. This study aims to determine the potential of compounds in sungkai plant leaves as immunostimulants. Isolate 1 was successfully isolated by vacuum chromatography to radial methods and characterized using UV-Vis spectroscopy, FT-IR, 1H-NMR, 13C-NMR, 2D-NMR (HMBC, HSQC, & COSY), and mass spectroscopy methods which showed that isolate 1 is a flavonoid compound namely apigenin. Then the apigenin compound that was successfully isolated was then tested for Granzyme, Interferon Gamma, and Perforin expression using 25 mice and divided into 5 groups where groups 2-5 were induced with the COVID-19 Vaccine to determine its activity as an immunostimulant, from the test indicate that apigenin with doses of 1 mg/kg BW, 25 mg/kgBW, and 50 mg/kgBW provide optimal effects to increase granzyme, Interferon Gamma, and Perforin levels in mice.
